Nowadays, speed and economy are the most important aspects in HPLC analyses as thousands batches are analyzed every month in various industries for quality control. Worldwide, the strong economic pressure of increasing price of many products necessitates the enhancement of analyses throughput in order to reduce manufacturing cost. The production cost can be reduced by reducing HPLC run time and low consumption of costly solvents. Recently, special type of superficially porous silica gel based columns of super fast speed and low solvent consumption have been introduced into the market, which are called new generation columns.

Chromanik Tech. Inc., Japan introduced superficially porous columns under the trade name of Sunshell with good peak shape, high stability and low bleeding. The chemistries of Sunshell columns include C8, C18, RP-aqua, Phenyl. Most recently, Chromanik Tech. Inc., Japan introduce Peptides and Proteins C18 based Wide pore (WP) C18 column with low carbon loading (HWP). The Sunshell sorbents comprises Non Porous Silica core with 1.5 µ of superficially porous RP bonded sorbent layer with narrow well controlled tighter Particle size distribution (dv 90/dv 10= 1.15); offering reinforced column efficiency comparable to sub 2 µ HPLC column performance. This unique configuration offers the performance advantages of sub-2µ particles with backpressures comparable to that of 3 µ Sorbent particles.

The technology embedded in Sunshell sorbent manufacturing consist of single step Co-acervation process resulting in superior homogenous superficially porous Sunshell sorbent unlike similar coating process involving multilayer superficially porous sorbent process. Such Multilayer Porous Coating  being Multi stage process results in Layer to layer diversity and non reproducibility of the particle size throughout the entire coating process. In regards to Sunshell the single step process delivers higher yields and more column to column reproducibility. 


Sunshell columns with Single step Superficial Coating along with unique HMODT Reverse phase bonding are capable of performance at wider pH range and temperature till 80 °C. Sunshell HPLC columns offer good peak shapes, high stability along with low bleeding and these columns can be used for UPLC, LC-MS as well as on conventional HPLC instruments. As of now in core shell category only Sunshell columns have proven stability data at 1% TFA (90 parts) pH 1 at 80 °C and at 10 mM sodium hydroxide (pH 10) at 50 °C.

Wishing Long Lives of Superficially Porous Particles HPLC Columns

1. Superficially Porous Particles HPLC Columns

Nowadays, scientists, academicians, industrialist and government authorities are looking for high speed analyses due to economic pressure all the world over. This requirement has been fulfilled by newly developed superficially porous particles columns (SPPCs). Generally, these particles have 2.7 and 1.7 µm as overall and central solid core diameters with a 0.50 µm outer thick porous layer with 90.0 Ǻ pores size. These porous silica gel particles are prepared by Fused Core Technology. The porous particles were prepared of various types of silica gels such as C8 and C18 and RP Amide and Hilic. The beauty of these columns is that in spite of small particle size they can be used with normal HPLC machine i.e. no need of costly UPLC. Presently, these types of columns are manufactured by Advanced Material Technology, USA (Halo), Supelco, USA (Express), Agilent, USA (Proshell 120), Phenomenex, Madrid Ave, Torrance, CA, USA  (Kinetex) and Chromanik Tech. Inc., Japan (Sunshell).

The pore size in thick porous layer is quite small and there is always risk of blockage of these columns resulting into an increase in back pressure during their uses. These columns are reversed phase and the separation mechanism is as usual of C18 columns i.e. hydrophobic interactions with stationary phase. The choice of mobile phases with these columns is buffers. The most commonly used buffers in HPLC are phospharte, acetate, formate, citrate, borate etc. HPLC separation is improved by using organic modifiers and most commonly used one is acetonitrile. Normally, the constituents of these buffers get precipitated by the addition of acetonitrile in buffers. This precipitation does not affect much in normal C18 columns due to large pore size but blocks superciliously porous silica gel particle based columns due to small pore size. Consequently, the back pressure starts to increase with respect to the operational time. It results in to the blockage of column after about 100 runs; depending on the mobile phase, duration of experiment and other experimental conditions. This results into a need of costly UPLC due to an increase in back pressure, poor reproducibility and finally the risk of column damage. During our experiments, we observed certain phenomenon and reached on some conclusions for reproducible results and long lives of these columns. Some most important precautions to achieve reproducible results and long lives are summarized below.

2. Precautions for Reproducible Results and Long Lives of SPPCs:

The precipitated buffer constituents appear to become trapped in small pore size of SPPCs. The following precautions should be taken when working these SPPCs:

2.1 The mobile phase should be prepared daily; especially for buffers having


2.1  The normal washing procedure is not sufficient to dissolve and remove the trapped buffer constituents. Therefore, a little rigorous washing is required to remove precipitated buffer ingredients. After the run of about 10-12 hrs time, the column should be washed with water at 0.5 mL/min. flow rate for 1.0 h followed by further washing at 1.0 mL/min at 1 h time.

2.3 Store column in water-acetonitrile (40:60, v/v) mobile phase.

3        Conclusion:

The reproducibility and life of a column is very important aspect in HPLC analyses. The reproducibility is the scientific requirement while life of column controls the economy of the method. Both are important in research and development and routine analyses in the industries. Therefore, the proposed precaution protocol is very important issue in SPPCs.
